چکیده
pBR322/Ultraviolet irradiation/Restriction endonuclease/Pyrimidine dimer Plasmid pBR322 DNA was irradiated with ultraviolet light at 254 nm and digested with various restric tion endonucleases. Analyses of the products suggested that the pyrimidine dimers which would have been formed just across the cutting site (Acc I), at the opposite position of the cutting site (Hind III, Eco RI, Bam HI), and at the position adjacent to the cutting site (Aha III) blocked the cleavage with the enzymes. In addition, experiments with Hind III, Eco RI and Bam HI showed that the extent of cleavage block appears to reflect the relative efficiency of dimer formation in sequences, TT, CT and CC.

متن کاملMapping the B-A conformational transition along plasmid DNA
A simple method is presented to monitor conformational isomerizations along genomic DNA. We illustrate properties of the method with the B-A conformational transition induced by ethanol in linearized pUC19 plasmid DNA. At various ethanol concentrations, the DNA was irradiated with ultraviolet light, transferred to a restriction endonuclease buffer and the irradiated DNA was cleaved by 17 restri...
متن کاملCleavage of mispaired heteroduplex DNA substrates by numerous restriction enzymes.
The utility of restriction endonucleases as a tool in molecular biology is in large part due to the high degree of specificity with which they cleave well-characterized DNA recognition sequences. The specificity of restriction endonucleases is not absolute, yet many commonly used assays of biological phenomena and contemporary molecular biology techniques rely on the premise that restriction en...
متن کاملCatalytic domain of restriction endonuclease BmrI as a cleavage module for engineering endonucleases with novel substrate specificities
Creating endonucleases with novel sequence specificities provides more possibilities to manipulate DNA. We have created a chimeric endonuclease (CH-endonuclease) consisting of the DNA cleavage domain of BmrI restriction endonuclease and C.BclI, a controller protein of the BclI restriction-modification system. The purified chimeric endonuclease, BmrI198-C.BclI, cleaves DNA at specific sites in t...
متن کاملDiversity of type II restriction endonucleases that require two DNA recognition sites.
Orthodox Type IIP restriction endonucleases, which are commonly used in molecular biological work, recognize a single palindromic DNA recognition sequence and cleave within or near this sequence. Several new studies have reported on structural and biochemical peculiarities of restriction endonucleases that differ from the orthodox in that they require two copies of a particular DNA recognition ...
